Abstract
In the central nervous system (CNS), the effective delivery of pharmacological agents remains limited by several brain barriers [i.e., the blood-brain barrier (BBB), blood-cerebrospinal fluid barrier (BCSFB), and blood-arachnoid barrier (BAB)]. In addition to these primary barriers, brain parenchymal cellular compartments can further regulate drug distribution in the CNS. This chapter provides an overview of the physiological properties of brain barriers (i.e., BBB, BCSFB, BAB), as well as the functional expression of drug transporters at these barriers and brain cellular compartments. Relevant drug transporters belonging to the ATP-binding cassette (ABC) and solute carrier (SLC) superfamilies in the CNS are discussed in detail. This chapter also describes the role of drug transporters in various CNS disorders (i.e., HIV-1 infection, epilepsy, cerebral folate deficiency disorders, and others) and addresses their contribution in the disposition of CNS pharmacotherapy.
