N2 - Understanding cell heterogeneity and protein expression levels at the single cell level is of high clinical and biological interest, but is rarely achieved with current technologies. Here, we show the detection and quantification of the apoptotic marker protein Bcl-2 from MCF-7 breast cancer cells with an integrated microfluidic device combined with MALDI mass spectrometry. We demonstrated effective lysis of cells on-chip and the capability of quantifying down to 2.63 x 1011 molecules of proteins from cells as an important milestone to further develop this approach into a protein single cell analysis platform.
